Help Starvation!!

Hi guys,
Have a 3B diesel and it's starting to play up. Engine is starved of fuel. Runs for a minute if I hand pump it then dies. Have changed fuel filter and lift pump as i had them to hand. No joy. Pressurised the fuel tank and she ran great for ten minutes. Immediately I went off the flat she stopped and is a POS to start again. Checked lines and no fuel in them. Thinking of dropping the tank to have a look. Is there a filter in there somewhere? Oh yep checked the lift pump in line filter that's good too. Any ideas?
Also have a Holley fuel pump to hand and possibility of fitting this but I would rather get to the real problem.
Any suggestions would be great.
