Running upside down for 5 seconds probably didn't hurt it. Typically it's the engine sitting upside down while you set up to recover it. Oil will sneak past the piston rings, and when you go to restart it you bend a connecting rod. That's why before restarting after a roll, or water ingestion you should always remove spark plugs or injectors/glow plugs and crank it to clear the cylinders of oil.
For diagnosing your problem I'm guessing you isolated it to number 1 by undoing each injector and looking for a change in engine note. You could do a compression check to be sure but removing a 12ht head is pretty easy, i'd just go ahead and do that. Probably worst case you need a few new conrods. Finding some may be difficult though because toyota no longer make parts for the 12ht. It won't hurt to throw in some new rings and bearings while your at it either.

well before i started it i pulled all 6 injectors out wound it ova for about 30 seconds took all the intake pipes off got the oil out of every thing but wat i cant figure out is when i left the fourby tracks it was on running on 5 i stoped at a mates joint for a bit and then when i left it was runing on 6 and wuld make a backfiring noise and go back and fourth from 5 to 6 and i have changed the pump and all 6 injectors i just dont no i havent pulled the motor apart yet becuse i am still doin the ute chop from the roll over but i have 2 blown up 12ht in my garage so if its just a rod then happy days
